Ondansetron, commonly known as Zofran, is a selective 5-HT3 receptor antagonist designed to prevent and treat nausea and vomiting. It works both centrally and peripherally by blocking serotonin receptors in the brain and gastrointestinal (GI) tract. Centrally, it targets receptors in the brain that trigger the sensation of nausea, while peripherally, it acts through the vagus nerve to address nausea and vomiting triggers in the GI system.
Ondansetron is most often used in situations where nausea and vomiting are common, such as before surgery, during radiation therapy, or alongside cancer chemotherapy. It is also effective in managing nausea associated with GLP-1 receptor agonist injections, such as semaglutide or tirzepatide, making it a valuable option for patients undergoing weight loss treatment.
